We constructed this device with BBa_J23119, BBa_I13504, BBa_J23119, BBa_K115002 and E1010. Means that it contains a GFP and a mCherry controlled by RNA thermometer. As thus, GFP and mCherry were expressed in a same cell and present wonderful color. Users can adjust the ratio between red and green through change of temperature. What's more, this device is designed for value the switch efficiency of RNA Thermometer: Heat stress will cause protein unfolding, entanglement, and unspecific aggregation, which can interfere with protein expression. Thus, the fluorescence difference is the combined effect of RNAT and heat stress. We use GFP to indicate factor caused by heat stress, RFP under the control of RNA thermometer to indicate the factor caused by heat stress and switch efficiency of RNA thermometer.

The Switch Efficiency Of RNA Thermometer Under Heat Stress
We value the circuits under 5 different temperatures: 28℃, 35℃, 37℃, 40℃ and 42℃.
We explored a new method to measure the switch efficiency: using 2 kinds of fluorescent proteins, a GFP indicate the heat stress, and a mCherry indicate the combined effect.
